ALP hydrolyses p-Nitrophenyl phosphatase into p-Nitrophenol and phosphate. The alkaline pH of the buffered medium, p-Nitrophenol is yellow. The color developed due to hydrolysis measured at 405 nm and is proportional to the Alkaline Phosphatase activity.
